目的:采用高分离度快速液相色谱法测定黄芪药材中毛蕊异黄酮的含量。方法:色谱柱:安捷伦Zorbax SB-C_(18)柱(50mm×4.6mm,1.7μm);流动相:0.1%甲酸水溶液(A)-乙腈(B),梯度洗脱,流速:0.6 ml·min~(-1);柱温:25℃;检测波长:280nm。结果:毛蕊异黄酮在6.05~121.00μg·ml~(-1)浓度范围内与峰面积呈良好线性(r=0.9999),平均回收率为96.17%(RSD=2.84%,n=6)。结论:该方法简便、快速、准确,重复性好,可用于黄芪药材的质量评价。
Objective: To determine the content of calycosin in Astragalus membranaceus by high performance liquid chromatography. Method: Column: Agilent Zorbax SB-C 18 column (50 mm × 4.6 mm, 1.7 μm); mobile phase: 0.1% formic acid in water ~ (-1); column temperature: 25 ℃; detection wavelength: 280nm. Results: There was a good linearity (r = 0.9999) for calycosin in the range of 6.05 ~ 121.00μg · ml ~ (-1) with an average recovery of 96.17% (RSD = 2.84%, n = 6). Conclusion: The method is simple, rapid, accurate and reproducible, and can be used for the quality evaluation of Radix Astragali.
