目的:建立HPLC法同时测定华山参滴丸中消旋山莨菪碱、东莨菪碱及阿托品的含量。方法:采用资生堂Capcell-Pak C18(4.6mm×250mm,5μm)色谱柱,以乙腈-醋酸盐缓冲液(5:95)为流动相A,乙腈-醋酸盐缓冲液(15:85)为流动相B,进行梯度洗脱(0~30min,60%A→35%A;30~40min,35%A→30%A;40~44min,30%A→20%A;44~45min,20%A→60%A;45~60min,60%A),流速1.0mL.min-1,检测波长210nm;柱温30℃。结果:消旋山莨菪碱、东莨菪碱及阿托品的线性范围分别为0.08~1.82,0.07~1.65,0.04~1.03μg(r=0.9999);平均回收率分别为99.1%(RSD=1.8%),97.0%(RSD=1.6%),104.0%(RSD=0.84%)。结论:所建立的HPLC法可同时测定华山参滴丸中消旋山莨菪碱、东莨菪碱及阿托品的含量。
Objective: To establish an HPLC method for the simultaneous determination of racimosan, scopolamine and atropine in Huashandishen dripping pills. Methods: The mobile phase A was acetonitrile-acetate buffer (5:95) and acetonitrile-acetate buffer (15:85) Mobile phase B, gradient elution (0-30 min, 60% A → 35% A; 30-40 min, 35% A → 30% A; 40-44 min, 30% A → 20% A; 44-45 min, 20 % A → 60% A; 45-60min, 60% A), flow rate 1.0mL.min-1, detection wavelength 210nm; Results: The linear ranges of racimustine, scopolamine and atropine were 0.08-1.82,0.07-1.65 and 0.04-1.03 μg, respectively (r = 0.9999). The average recoveries were 99.1% (RSD = 1.8%) and 97.0% (RSD = 1.6%), 104.0% (RSD = 0.84%). Conclusion: The established HPLC method can simultaneously determine the content of racimustine, scopolamine and atropine in Huashandishen dripping pills.
