[目的 ]建立一种简便有效的体外微量测定方法 ,以监测恶性疟原虫对咯萘啶 /磺胺多辛 /乙胺嘧啶敏感性的变化。 [方法 ]设计制作咯萘啶 /磺胺多辛 /乙胺嘧啶 3种药物伍用测定板 ,采用WHO推荐的Rieckmann体外微量法 (WHO标准化法 )测定涂药板效果。测得的原始数据采用W H Wernsdorfer的专门软件进行分析 ,判定用体外微量法测定恶性疟原虫对上述 3种药物伍用测定板敏感性的可行性 ,并依据分数抑制浓度 (FIC)判定咯萘啶与磺胺多辛 /乙胺嘧啶之间是否有协同效应。 [结果 ]3药伍用测定板效果稳定 ,其ED5 0 分别为咯萘啶 0 11pmol/井、磺胺多辛 2 15 12pmol/井和乙胺嘧啶 2 6 9pmol/井。同时证明 ,咯萘啶与磺胺多辛 /乙胺嘧啶之间有协同效应。 [结论 ]恶性疟原虫对咯萘啶 /磺胺多辛 /乙胺嘧啶的敏感性可用体外微量法进行测定。
[Objective] To establish a simple and effective in vitro micro-assay to monitor the change of susceptibility to pyronaridine / sulfadoxine / pyrimethamine in Plasmodium falciparum. [Method] We designed and prepared three kinds of drugs, pyronaridine / sulfadoxine / pyrimethamine, and measured the effect of coated tablets by Rieckmann in vitro micro-method (WHO standardization method) recommended by the WHO. The measured raw data was analyzed by WH Wernsdorfer’s special software to determine the feasibility of determining the susceptibility of Plasmodium falciparum to the above three drugs by in vitro microdilution and to determine pyronaridine according to fractional inhibitory concentration (FIC) And sulfadoxine / pyrimethamine whether there is a synergistic effect. [Results] The results of the three-drug assay plate were stable with ED50 values of pyronaridine 0 11 pmol / well, sulfadoxine 2 15 12 pmol / well and pyrimethamine 269 pmol / well. At the same time, it is proved that there is a synergistic effect between pyronaridine and sulfadoxine / pyrimethamine. [Conclusion] The sensitivity of Plasmodium falciparum to pyronaridine / sulfadoxine / pyrimethamine can be determined by in vitro microassay.
