In 1984, Heyendran et al. Used 150 ml permeate (mOsm) of fructose, sodium citrate and hypotonic solution with ionic strength of 0.15 as the medium of semen samples to evaluate the function and structure of the sperm membrane under the condition of sperm swelling. Because sperm tail is particularly sensitive to hypotonic conditions, a typical change in sperm morphology can be observed with a phase contrast microscope. A simple method is to use swollen water (a very strong hypotonic fluid) as a hypotonic medium to swollen sperm to evaluate the integrity of the membrane, ie, water experiments.
