以直链淀粉为包合载体,采用密封控温包合技术制备异维A酸包合物,通过粉末X-射线衍射及DSC测试验证包合物的形成。运用动力学原理建立异维A酸/直链淀粉包合物的酶控释放动力学方程,通过方程推断在α-淀粉酶作用下药物释放的动力学规律。结果表明,在无α-淀粉酶存在时,药物释放符合一级动力学模型;当α-淀粉酶存在时,药物释放是由脱包和酶解共同作用的加速过程。预示异维A酸/直链淀粉包合物可通过加入α-淀粉酶调控药物的释放。
The inclusion complex of amylose was used to prepare isotretinoin inclusion complex by sealed temperature-controlled inclusion technique. The formation of inclusion complex was confirmed by powder X-ray diffraction and DSC. The kinetic principle was used to establish the kinetic equation for the controlled release of isotretinoin / amylose inclusion complex, and the kinetics of drug release under the action of α-amylase was deduced through the equation. The results showed that in the absence of α-amylase, the drug release was in accordance with the first-order kinetic model; in the presence of α-amylase, the drug release was accelerated by the combination of unpacking and enzymolysis. It is indicated that isotretinoin / amylose inclusion complex can regulate drug release by adding alpha-amylase.
