为研究ALAD基因多态性对铅致神经毒性的影响 ,对 179名生活在严重铅污染环境中的 6~ 8岁儿童进行基因型分析、血铅和锌原卟啉 ,身高、体重、血红蛋白、智商和反应时测定。结果表明 ,与ALAD1- 1纯合子相比 ,ALAD1- 2杂合子的血铅和锌原卟啉都轻度增高但没达到显著性水平。而两组的体格和智力发育参数都十分接近 ,但ALAD1- 2组的反应时延长 (P =0 0 2 )。在应用协方差分析充分均衡了两组的血铅水平后 ,ALAD1- 2的选择反应时间与ALAD1- 1组十分接近。结果提示ALAD1- 2基因型对铅致神经毒性效应与AL AD1- 1的效应存在差异 ,其机理是基因变异介导的体内铅负荷增高所致
In order to study the effect of ALAD gene polymorphisms on lead-induced neurotoxicity, genotypes were analyzed in 179 children aged 6-8 years who lived in serious lead-contaminated environment. Blood lead, zinc protoporphyrin, height, body weight, hemoglobin, IQ and reaction time measurements. The results showed that the blood lead and zinc protoporphyrin of ALAD1-2 heterozygote were mildly elevated but did not reach significance level compared with ALAD1-1 homozygote. The physical and mental development parameters of both groups were very close, but the response time of ALAD1-2 group was prolonged (P = 0 0 2). After applying the analysis of covariance to fully equalize the blood lead level of both groups, the ALAD1-2 selective response time was very similar to ALAD1-1. The results suggest that the ALAD1-2 genotype has a different effect on the lead-induced neurotoxic effect and the AL-AD1-1 effect by a mechanism of gene mutation-mediated increase of lead load in the body
