目的观察双氢青蒿素对Balb/c小鼠非特异以及特异性免疫功能的影响。方法采用经口方式给药,设31.25、62.5、125和250 mg/kg 4个剂量组,并设溶剂对照组,时间两周。观察双氢青蒿素对淋巴器官脏器湿重以及脏器系数的影响,采用乳酸脱氢酶释放法检测NK细胞活性,流式细胞技术测定巨噬细胞吞噬功能,溶血空斑实验检测对体液免疫功能,迟发型变态反应检测细胞免疫功能以及刀豆蛋白A活化的淋巴细胞转化实验(MTT法)检测T细胞的增殖功能。结果在31.25~250 mg/kg,双氢青蒿素能够增强NK细胞活性,降低巨噬细胞的吞噬功能,在250 mg/kg剂量下,能够抑制迟发型变态反应,并且抑制非活化的T淋巴细胞的增殖,但是对刀豆蛋白活化的淋巴细胞增殖没有显著影响,对体液免疫功能没有显著性影响。结论双氢青蒿素能够抑制迟发型变态反应,并且抑制非活化的T淋巴细胞增殖;降低巨噬细胞的吞噬功能,提高NK细胞活性;但是对体液免疫没有显著影响。
Objective To observe the effect of dihydroartemisinin on non-specific and specific immune function in Balb / c mice. Methods By oral administration, four doses of 31.25, 62.5, 125 and 250 mg / kg were given, and the solvent control group was given for two weeks. The effect of dihydroartemisinin on the wet weight and organ coefficient of lymphoid organs was observed. The activity of NK cells was detected by lactate dehydrogenase release assay. The phagocytosis of macrophages was determined by flow cytometry. Immune function, delayed type hypersensitivity test of cellular immune function and Concanavalin A-activated lymphocyte transformation assay (MTT method) were used to detect the proliferation of T cells. Results At 31.25-250 mg / kg, dihydroartemisinin enhanced NK cell activity and decreased macrophage phagocytosis. At 250 mg / kg, dihydroartemisinin was able to inhibit the delayed-type hypersensitivity and inhibit non-activated T lymphocytes Cell proliferation, but had no significant effect on the proliferation of cytoplasm-activated lymphocytes and had no significant effect on humoral immune function. Conclusions Dihydroartemisinin can inhibit the delayed-type hypersensitivity, inhibit the proliferation of non-activated T lymphocytes, decrease the phagocytosis of macrophages and increase the activity of NK cells. However, it has no significant effect on humoral immunity.
