目的:探讨补益脾肾方对阿霉素肾病大鼠肾间质转化生长因子-β1(TGF-β1)和结缔组织生长因子(CT-GF)表达的影响。方法:将32只雄SD大鼠随机分为空白组、阿霉素肾病模型组、贝那普利治疗组、补益脾肾中药治疗组。采用一次性尾静脉注射阿霉素建立阿霉素肾病模型,于造模后第2周对阿霉素肾病大鼠分别用贝那普利和中药治疗。于造模后第6周处死所有大鼠,切片做Masson染色观察肾小管及间质的病理变化;应用免疫组织化学方法检测肾组织中TGF-β1、CTGF的蛋白质表达。结果:补益脾肾法可以下调阿霉素肾病大鼠肾间质TGF-β1及CTGF的表达(P<0.05),延缓间质纤维化的进展。结论:中医补益脾肾方可以通过下调肾间质TGF-β1及CTGF的表达,延缓阿霉素肾病大鼠肾小管间质纤维化的进展。
Objective: To investigate the effect of replenishing spleen-invigorating recipe on the expression of transforming growth factor-β1 (TGF-β1) and connective tissue growth factor (CT-GF) in adriamycin-induced nephropathy rats. Methods: Thirty-two male SD rats were randomly divided into blank control group, adriamycin nephropathy model group, benazepril treatment group and Chinese herbal medicine for treating spleen and kidney. One-time intravenous injection of doxorubicin was used to establish the model of adriamycin nephropathy. Rats in adriamycin-induced nephropathy were treated with benazepril and Chinese herbs respectively in the second week after the model was established. All the rats were sacrificed at the 6th week after the model establishment. Masson staining was used to observe the pathological changes of tubules and interstitium. The protein expression of TGF-β1 and CTGF in renal tissues was detected by immunohistochemistry. Results: The recipe for invigorating the spleen and kidney can down-regulate the expression of TGF-β1 and CTGF in the renal interstitium of adriamycin-induced nephropathy rats (P <0.05), delay the progression of interstitial fibrosis. Conclusion: Chinese herbs for benefiting spleen and kidney can delay the progress of tubulointerstitial fibrosis in rats with adriamycin-induced nephropathy by down-regulating the expression of TGF-β1 and CTGF in the renal interstitium.
