目的:建立鸦胆子油的HPLC-ELSD指纹图谱分析方法,并与掺伪鸦胆子油指纹图谱进行比较。方法:运用SSI高效液相色谱仪和奥泰2000蒸发光检测器,采用Waters C_(18)(200 mm×4.6 mm,5μm)色谱柱,流动相:乙腈-二氯甲烷(60∶40),流速:1.2 ml·min~(-1),检测温度:85℃,柱温:40℃,进样量:15μl。结果:方法的精密度、重复性与稳定性良好,相对保留时间和相对峰面积的RSD均<3.0%,各峰分离度良好,指纹信息完整,符合指纹图谱技术规范。在此方法的基础上,建立了鸦胆子油及掺伪鸦胆子油HPLC-ELSD指纹图谱,并将相似度进行比较。结论:本试验为鸦胆子油质量控制和市场监管提供了重要依据。
OBJECTIVE: To establish HPLC-ELSD fingerprinting method of Brucea javanica oil and compare with fingerprints of Brucea javanica oil. Methods: A Waters C18 (200 mm × 4.6 mm, 5 μm) column was used with a mobile phase of acetonitrile-methylene chloride (60:40) using an SSI high performance liquid chromatograph and an Altai 2000 evaporative detector. Flow rate: 1.2 ml · min -1, detection temperature: 85 ℃, column temperature: 40 ℃, injection volume: 15μl. Results: The precision, reproducibility and stability of the method were good. The RSDs of relative retention time and relative peak area were both less than 3.0%. The peak separation was good and fingerprint information was complete. Based on this method, HPLC-ELSD fingerprints of Brucea javanica oil and Brukeria javanica were established, and the similarities were compared. Conclusion: This experiment provides an important basis for the quality control and market supervision of Brucea javanica oil.
