目的建立湿生扁蕾药材中木犀草素和1,7-二羟基-3,8-二甲氧基酮的含量测定方法。方法采用反相高效液相色谱法(RP-HPLC),色谱柱为ZORBAXSB-C18(4.6mm×250mm,5μm);流动相为甲醇(A)-0.4%磷酸水溶液(B)梯度洗脱,0~15min,A∶B=50∶50,15~60min,A∶B=55∶45;检测波长260nm;流速1mL/min;柱温30℃。结果1,7-二羟基-3,8-二甲氧酮和木犀草素分别在0.084~0.84μg和0.184~1.84μg范围内线性关系良好,相关系数分别为0.9994和0.9997;平均回收率分别为96.82%和97.19%,RSD均小于3%。结论该方法简便准确,重现行好,可作为湿生扁蕾药材的质量控制方法。
Objective To establish a method for the determination of luteolin and 1,7-dihydroxy-3,8-dimethoxyketone in wet buds. Methods The reversed-phase high performance liquid chromatography (RP-HPLC) was used. The column was ZORBAXSB-C18(4.6mm×250mm, 5μm). The mobile phase was gradient elution of methanol(A)-0.4% phosphoric acid aqueous solution(B). -15 min, A:B = 50:50, 15 - 60 min, A:B = 55:45; detection wavelength 260 nm; flow rate 1 mL/min; column temperature 30C. Results The linearity of 1,7-dihydroxy-3,8-dimethoxone and luteolin in the range of 0.084-0.84 μg and 0.184-1.84 μg were good, and the correlation coefficients were 0.9994 and 0.9997, respectively; the average recoveries were 96.82% and 97.19%, RSD were less than 3%. Conclusion The method is simple and accurate, and it can be used as a method for quality control of wet buds.
