目的探讨反义寡核苷酸(ASON)特异性抗病毒作用。方法在HepG22.2.15细胞中观察了4段16聚针对乙型肝炎病毒(HBV)基因不同功能区的ASON对病毒复制的抑制作用。结果ASON能显著抑制HBV基因的抗原表达(P<0.001),在S基因区设计的ASON对HBsAg的抑制作用明显优于C基因区的ASON(P<0.05)。反之,在C基因区设计的ASON对HBeAg的抑制作用又明显优于S和Pre-S2基因区的ASON(P<0.01)。4段ASON的联合用药并不能增强其对HBV的抑制作用。结论ASON可能成为一种有开发潜力的抗病毒药物。
Objective To investigate antisense oligonucleotide (ASON) specific antiviral effect. Methods In HepG22.2.15 cells, we observed the inhibitory effect of ASON on viral replication in four segments of 16-poly-AS against different functional regions of hepatitis B virus (HBV) gene. Results ASON could significantly inhibit the expression of HBV antigens (P <0.001). The inhibitory effect of ASON on S gene was better than that of C genes (P <0.05). Conversely, the inhibitory effect of ASON designed in the C gene region on HBeAg was significantly better than that of ASON in the S and Pre-S2 gene regions (P <0.01). 4-ASON combination does not enhance its inhibitory effect on HBV. Conclusion ASON may become a development potential of antiviral drugs.
