目的比较白木香结香前后内生真菌群落结构和变化,探讨内生真菌对白木香结香的影响。方法分别提取白木香结香前后总DNA,采用真核生物r DNA ITS区通用引物ITS1F-gc/ITS2进行扩增,变性梯度凝胶电泳(Polymerase Chain Reaction-Denaturing Gradient Gel Electrophoresis,PCR-DGGE)分离后,切割回收条带进行克隆、测序和比对。结果 PCR-DGGE图谱聚类表明,未结香组与结香组分别聚在一起;与未结香组比较,结香组基因型丰富度、香农多样性指数显著提高(P<0.01);回收条带测序比对结果多为真菌,极个别序列无比对结果。结论白木香结香前后内生真菌群落存在差异,内生真菌可能参与白木香的结香过程。
Objective To compare the structure and changes of endophytic fungi communities before and after the fruiting bodies of Baimu Xiang and explore the effect of endophytic fungi on the fruiting bodies of white wood. Methods The total DNA was extracted before and after the A. officinalis, and ITS1F-gc / ITS2 was amplified by polymerase chain reaction-denaturing gradient gel electrophoresis (PCR-DGGE) After recovery, the bands were excised for cloning, sequencing and alignment. Results The cluster analysis by PCR-DGGE showed that the unjumbar group and the joss-stick group were clustered together. Compared with the unjumbar group, the genotype abundance and Shannon’s diversity index increased significantly (P <0.01) Band sequencing comparison results are mostly fungi, very few individual sequence incomparable results. Conclusion There are differences in endophytic fungi communities before and after the end of the fruiting of white wood, and the endophytic fungi may be involved in the process of the end of fragrant wood.
