[目的]研究香烟提取物(CSE)对人非小细胞肺癌(NSCLC)细胞生物学行为的影响及可能机制。[方法]CSE处理NSCLC细胞系A549、LH7与BE1,对照组正常培养。Transwell实验检测两组细胞的侵袭能力,RT-PCR法和Westernblot法检测两组细胞MTA1mRNA及蛋白表达情况。[结果]CSE处理各CSE组细胞侵袭能力增加,穿过膜细胞数明显高于对照组;且各CSE组中MTA1mRNA和蛋白表达水平也明显高于对照组(P<0.05)。[结论]香烟提取物导致NSCLC细胞系侵袭能力增加与其促进NSCLC细胞系中MTA1mRNA及蛋白表达有关。
[Objective] To investigate the effect of cigarette smoke extract (CSE) on the biological behavior of human non-small cell lung cancer (NSCLC) cells and its possible mechanism. [Method] NSCLC cell line A549, LH7 and BE1 were treated with CSE and the control group was cultured normally. The invasion ability of the two groups of cells was detected by Transwell assay. The expression of MTA1 mRNA and protein was detected by RT-PCR and Western blot. [Results] The invasive ability of CSE cells in each CSE group was increased and the number of cells passing through the membrane was significantly higher than that in the control group. The expression of MTA1 mRNA and protein in CSE group was also significantly higher than that in the control group (P <0.05). [Conclusion] The increase of invasiveness of NSCLC cell lines caused by cigarette extract is related to the promotion of MTA1 mRNA and protein expression in NSCLC cell lines.
