目的本研究主要探讨HAS-2沉默前后对大肠癌细胞上皮间质转化(EMT)的影响。方法分别用Realtime PCR及Western blot方法检测HAS-2 siRNA转染前后E-cadherin、Vimentin、Slug、Snail mRNA、蛋白表达水平变化,用免疫荧光法观察HAS-2 siRNA转染前后E-Cadherin、Vimentin表达的变化。结果 E-Cadherin蛋白和mRNA在大肠癌细胞的表达水平在HAS-2 siRNA转染组比阴性对照组明显上调(<0.05),而Vimentin蛋白和mRNA,Snail mRNA and Slug蛋白在大肠癌细胞的表达水平在HAS-2 siRNA转染组表达水平比阴性对照组明显下调(<0.05)。结论 HAS-2沉默对大肠癌细胞EMT有抑制作用。
Objective To investigate the effect of HAS-2 silencing on the epithelial-mesenchymal transition (EMT) in colorectal cancer cells. Methods The mRNA and protein expression of E-cadherin, Vimentin, Slug and Snail before and after HAS-2 siRNA transfection were detected by Realtime PCR and Western blot respectively. The expression of E-Cadherin, Vimentin Changes in expression. Results The expression of E-Cadherin protein and mRNA in colorectal cancer cells was significantly upregulated in HAS-2 siRNA transfected group (P <0.05), while the expression of Vimentin protein, Snail mRNA and Slug protein in colorectal cancer cells The level of HAS-2 siRNA transfection group expression level was significantly lower than the negative control group (<0.05). Conclusion HAS-2 silencing can inhibit the EMT of colorectal cancer cells.
