目的 为了探讨小儿颅内肿瘤化疗失败的原因 ,本实验对 2 0例小儿颅内肿瘤P 糖蛋白的表达情况进行研究。方法 用单克隆抗体JSB1及免疫组化法检测P 糖蛋白。结果 小儿颅内肿瘤P 糖蛋白表达很低 ,2 0例中 8例表达于血管内皮细胞 ,7例表达于纤维细胞 ,5例表达于浸润的淋巴细胞 ,肿瘤细胞只 1例表达。结论 小儿颅内肿瘤化疗失败的原因是患儿血脑屏障P 糖蛋白表达 ,表现为对抗癌药物较强耐受而非肿瘤细胞表达P 糖蛋白引起耐药 ,这种差别有显著的意义。需设计新的化疗途径克服这一障碍
Objective To investigate the causes of failure of intracranial tumors in children with chemotherapy, the present study was to investigate the expression of P-glycoprotein in 20 children with intracranial tumors. Methods Monoclonal antibody JSB1 and immunohistochemistry were used to detect P glycoprotein. Results The expression of P-glycoprotein in children with intracranial tumors was very low. Among 20 cases, 8 were expressed in vascular endothelial cells, 7 in fibroblasts, 5 in infiltrating lymphocytes and 1 in tumor cells. Conclusion The reason for the failure of chemotherapy in pediatric intracranial tumors is the expression of P-glycoprotein in the blood-brain barrier in children. The difference is significant because it is more resistant to anticancer drugs than to P-glycoprotein induced by tumor cells. Design new chemotherapeutic approaches to overcome this barrier
