大鼠孕19~21天的胚胎小脑,分别放在4℃、培养液1640及-15℃加有10%DMSO抗冻剂的培养液1640中。两组材料保存1、3、7天后。分别移植于成年同种大鼠创伤小脑内。均在移植后12天取受体小脑行常规组织学切片观察。结果是移植的脑组织大部分成活,其组织层次及细胞结构正常、清晰、且有细胞分裂。在-15℃、保存7天的胚脑组织移植后脑细胞均变性死亡。表明4℃是短期保存移植用胚脑组织的最适温度;移植用的胚脑组织低温保存是一可行和适用的方法。
Rat embryos 19 to 21 days of gestation, respectively, at 4 ℃, 1640 medium and -15 ℃ plus 10% DMSO antifreeze medium 1640. Two groups of materials were stored 1,3,7 days later. Were transplanted in adult traumatic rat cerebellum. 12 days after transplantation, the acceptor cerebellum was examined by routine histological sections. The result is that most of the transplanted brain tissue survives, with normal tissue structure and cell structure, with clear cell division. Brain cells degenerated after embryo brain transplantation at -15 ℃ for 7 days. The results showed that 4 ℃ is the optimum temperature for short term preservation of embryonic brain tissue for transplantation. Cryopreservation of embryonic brain tissue for transplantation is a feasible and applicable method.
