综合利用硅胶、ODS及正相、半制备高效液相等多种色谱技术,对蟾皮大孔树脂15%乙醇洗脱部位进行化学成分分离,运用现代波谱学方法结合文献对分离得到的化合物进行结构鉴定。该研究从蟾皮中共分离鉴定了15个亲水性化合物,分别为:4,5-dimethyl-1,3,4,5-tetrahydropyrrolo[4,3,2-de]quinolin-6-ol(1),5-羟色胺(2),N-methyl serotonin(3),蟾蜍素(4),1,2,3,4-tetrahydro-6-hydroxy-β-carboline(5),5-甲氧基色胺(6),甜菜碱(7),咖啡因(8),蟾毒色胺(9),shepherdine(10),色氨酸(11),5-羧基吲哚-3-乙酸(12),5-羟基色醇(13),2-methyl-6-hydroxy-1,2,3,4-tetrahydro-β-carboline(14),蟾蜍噻咛(15)。其中化合物1为新化合物,化合物5为新天然产物并首次报道其碳谱,化合物4~8,10~14为首次从蟾皮中分离得到。
The chemical components of 15% ethanol elution fractions of toad skin macroporous resin were separated by silica gel, ODS and many kinds of chromatographic techniques, such as normal phase and semi-preparative high performance liquid chromatography. The compounds were isolated by modern spectroscopy and literature Structure Identification. In this study, fifteen hydrophilic compounds were isolated and purified from the skin of toad: 15,4-dimethyl-1,3,4,5-tetrahydropyrrolo [4,3,2-de] quinolin- ), Serotonin (3), toadin (4), 1,2,3,4-tetrahydro-6-hydroxy-β-carboline Betaine (6) betaine (7) caffeine (8) bufotramine (9) shepherdine (10) tryptophan (11) - Hydroxycholesterol (13), 2-methyl-6-hydroxy-1,2,3,4-tetrahydro-β-carboline (14), Toadia thiazole (15). Among them, compound 1 was a new compound, compound 5 was a new natural product and its carbon spectrum was first reported. Compounds 4 ~ 8 and 10 ~ 14 were isolated from toad for the first time.
