本文研究了各种影响酶促反应速度的因素对从猪、牛肾中分离得到的氨基酰化酶拆分DL-Ala制备D-Ala的影响,结果表明,酶促反应的最适条件为:N-乙酰-DL-Ala的浓度为0.2mol/L,酶用量700u/mmol底物,反应温度37℃,pH7.0,反应时间5h。牛肾氨基酰化酶和猪肾氨基酰化酶的拆分率和D-Ala得率最高分别为90%和84%;89.7%和66.1%,表明牛肾氨基酰化酶的拆分率和得率均优于猪肾氨基酰化酶。
In this paper, we studied the effects of various factors affecting the rate of enzymatic reaction on the separation of D-Ala from DL-Ala by aminoacylase isolated from porcine and bovine kidney. The results showed that the optimal conditions for enzymatic reaction were as follows: The concentration of N-acetyl-DL-Ala was 0.2mol / L, enzyme dosage 700u / mmol substrate, the reaction temperature 37 ℃, pH7.0, reaction time 5h. Bovine kidney amino acylase and porcine aminoacylase resolution and D-Ala yield the highest were 90% and 84%; 89.7% and 66.1%, indicating that bovine kidney amino acylase Resolution and yield are better than porcine renal amino acylase.
