目的探讨β-catenin、Smad4在胰腺癌发生、发展、转移、浸润过程中的作用。方法应用免疫组化方法检测β-catenin、Smad4在正常胰腺、胰腺癌组织中的表达。结果β-catenin在胰腺癌中的异常表达率为68.33%,明显高于正常组织(0),异常表达与年龄、性别、病理分化程度无关;与有无淋巴结转移、临床分期相关,有淋巴结转移的异常表达率明显高于无淋巴结转移,Ⅲ+Ⅳ期的异常表达率明显高于Ⅰ+Ⅱ期。Smad4在胰腺癌的阳性表达(33/60)明显低于正常组织(17/20),阳性表达与年龄、性别、病理分化程度、有无淋巴结转移、临床分期无相关性;在胰腺癌中β-catenin的异常表达与Smad4的阳性表达相关。结论β-catenin、Smad4共同参与了胰腺癌的发生,wnt/β-catenin通路和TGF-β/Smad通路可能存在交汇点。
Objective To investigate the role of β-catenin and Smad4 in the development, metastasis and invasion of pancreatic cancer. Methods The expressions of β-catenin and Smad4 in normal pancreas and pancreatic cancer tissues were detected by immunohistochemistry. Results The abnormal expression rate of β-catenin in pancreatic cancer was 68.33%, which was significantly higher than that in normal tissues (0). The abnormal expression was not related to age, sex and pathological differentiation. There was correlation between the expression of β-catenin and lymph node metastasis Was significantly higher than that without lymph node metastasis. The abnormal expression rate in stage Ⅲ + Ⅳ was significantly higher than that in stage Ⅰ + Ⅱ. The positive expression of Smad4 in pancreatic cancer (33/60) was significantly lower than that in normal tissues (17/20). There was no correlation between the positive expression of Smad4 and age, sex, pathological differentiation, lymph node metastasis and clinical stage. The abnormal expression of -catenin is correlated with the positive expression of Smad4. Conclusions β-catenin and Smad4 are involved in the carcinogenesis of pancreatic cancer. The junction of wnt / β-catenin pathway and TGF-β / Smad pathway may exist.
