目的:观察山楂叶总黄酮对前脂肪细胞3T3-L1增殖、分化和分泌Leptin、PAI-1的影响,探讨其调脂作用的可能机制。方法:培养3T3-L1细胞,并用不同浓度的山楂叶总黄酮进行干预,以四甲基偶氮唑盐(MTT)法检测细胞增殖,用油红O染色和染色比色法分析脂肪细胞的分化程度。用ELISA法检测细胞培养上清中Leptin、PAI-1的含量。结果:山楂叶总黄酮中浓度组能促进前脂肪细胞的增殖,但抑制其向成熟脂肪细胞分化,山楂叶总黄酮对成熟脂肪细胞分泌Leptin、PAI-1有明显抑制作用,且呈剂量依赖性。结论:山楂叶总黄酮可能通过抑制前脂肪细胞分化和影响其分泌因子而达到调脂作用。
Objective: To observe the effects of hawthorn leaves flavonoids on the proliferation, differentiation and secretion of leptin and PAI-1 in 3T3-L1 cells, and to explore the possible mechanism of fat regurgitation. METHODS: 3T3-L1 cells were cultured and treated with different concentrations of total flavonoids in Hawthorn leaves. The proliferation of cells was detected by MTT assay. Results: The concentration of total flavonoids in hawthorn leaves promoted the proliferation of preadipocytes, but inhibited its differentiation into mature adipocytes. Hawthorn leaf flavonoids significantly inhibited the secretion of Leptin and PAI-1 from mature adipocytes in a dose-dependent manner. . Conclusion: The total flavonoids of hawthorn leaves may regulate fat by inhibiting the differentiation of preadipocytes and influencing the secretory factors.
