目的探讨参芎化瘀胶囊对脑缺血神经功能损伤的影响。方法 40只雄性SD大鼠分成假手术组、模型组、参芎化瘀胶囊高、低剂量组,采用改良的Pulsineli 4血管阻断(4-VO)法制作全脑缺血模型,脑缺血48 h,苏木精-伊红(HE)染色观察海马区神经细胞形态变化,原位缺口末端标记(TUNEL)法检测凋亡细胞,八臂迷宫法测试动物学习记忆功能,转棒法测试动物综合运动功能。结果与假手术组比较,模型组凋亡神经细胞数量(33.45±3.48)增多,动物进入迷宫总错误次数(8.94±3.78)、重复错误次数(7.32±2.80)、工作记忆错误次数(3.48±1.26)增多,综合运动评分(79.1±24.5)降低;与模型组比较,参芎化瘀胶囊组凋亡神经细胞数量(17.47±1.84)减少,动物进入迷宫总错误次数(3.00±1.80)、重复错误次数(2.10±0.85)降低,综合运动功能评分(208.9±32.5)提高。结论参芎化瘀胶囊可抑制全脑缺血大鼠神经细胞凋亡,改善脑缺血后神经功能损伤。
Objective To investigate the effect of Shenxionghuayu Capsule on nerve function damage after cerebral ischemia. Methods Forty male Sprague-Dawley rats were divided into sham operation group, model group and Shenxionghuayu capsule high and low dose groups. The model of global cerebral ischemia was established by modified 4-VO (Pulsineli 4) 48 h. The morphological changes of hippocampal neurons were observed by hematoxylin and eosin (HE) staining. Apoptotic cells were detected by TUNEL method, animal learning and memory function by eight-arm maze method, Comprehensive motor function. Results Compared with the sham operation group, the number of apoptotic neurons in the model group increased (33.45 ± 3.48), the total number of errors in the maze (8.94 ± 3.78), the number of repeated errors (7.32 ± 2.80), the number of working memory errors (3.48 ± 1.26 (79.1 ± 24.5). Compared with the model group, the number of apoptotic nerve cells in Shenxionghuayu Capsule group decreased (17.47 ± 1.84), the total number of errors in animals entering the maze was (3.00 ± 1.80), repeated errors (2.10 ± 0.85) decreased, and comprehensive motor function score (208.9 ± 32.5) increased. Conclusion Shenxionghuayu capsule can inhibit neuronal apoptosis in rats with global cerebral ischemia and improve neurological impairment after cerebral ischemia.
