目的了解山东省不同地区淡色库蚊成蚊对溴氰菊酯抗性与kdr等位基因的突变关联情况。方法采用ASPCR(allelespecificPCR)技术,检测不同种群淡色库蚊成蚊的kdr等位基因频率。结果成蚊接触筒检测济南、济宁和青岛地区的淡色库蚊雌蚊,其死亡率分别为39.62%、37.25%和83.81%;通过回归分析ASPCR检测的kdr等位基因频率结果,显示不同地区淡色库蚊对溴氰菊酯抗性L1014F型和L1014S型突变表型与kdr等位基因频率有明显的相关性。结论淡色库蚊kdr基因突变与溴氰菊酯抗药性表型呈正相关,ASPCR方法对于检测kdr等位基因的可行性亟待验证性研究。
Objective To understand the relationship between resistance to deltamethrin and the kdr allele in adult mosquito Culex pipiens pallens in different areas of Shandong Province. Methods ASPCR (allelespecific PCR) was used to detect the kdr allele frequency of adults of Culex pipiens pallens from different populations. Results The adult mosquitoes were negative for 39.62%, 37.25% and 83.81% of the mosquitoes in Jinan, Jining and Qingdao respectively. The frequency of kdr alleles detected by ASPCR was analyzed by regression analysis. Culex pipiens quinquefasciatus deltamethrin resistant L1014F and L1014S mutant phenotype and kdr allele frequency was significantly correlated. Conclusion The mutation of kdr gene in Culex pipiens pallens is positively correlated with the resistance phenotype of deltamethrin. The feasibility of ASPCR method in detecting kdr allele is urgent to be verified.
