脓毒症早期大鼠脑线粒体损伤的研究

目的:探索脓毒症早期脑细胞线粒体功能及结构损伤情况。方法:清洁级雄性SD大鼠40只,以抽签方式随机分为4组:正常对照组,3h脓毒症组(3LPS组),6h脓毒症组(6LPS组)和24h脓毒症组(24LPS组),每组10只。腹腔注射革兰阴性菌脂多糖(LPS)溶液建立脓毒症大鼠动物模型,各脓毒症组大鼠在相应时点处死后分离获得线粒体。流式细胞仪测定各组大鼠脑线粒体膜电位、电镜观察各种神经细胞线粒体的结构损伤。结果:3LPS组大鼠脑线粒体膜电位较正常对照组明显降低[(0.5±0.5)vs(1.2±0.6),P<0.05];6LPS组和24LPS组大鼠脑线粒体膜电位与正常对照组无明显差异[(1.4±0.7)vs(1.2±0.6),(1.6±0.7)vs(1.2±0.6)]。6LPS组大鼠神经元及星形胶质细胞线粒体超微结构评分较对照组明显升高(P=0.000),脓毒症各组小胶质细胞和少突胶质细胞线粒体超微结构评分较对照组无明显差异。结论:脓毒症早期,大鼠脑组织线粒体存在可逆性功能和结构损伤,功能损伤先于结构损伤出现;脓毒症大鼠神经元细胞和星形胶质细胞线粒体损伤较为明显,而小胶质细胞和少突胶质细胞具有较强的抗损伤能力。 Objective: To explore the early mitochondrial function and structural damage of brain cells in sepsis. Methods: Forty clean male Sprague-Dawley rats were randomized into 4 groups by random sampling: normal control group, 3h sepsis group (3LPS group), 6h sepsis group (6LPS group) and 24h sepsis group 24 LPS group), 10 in each group. The animal model of sepsis was established by intraperitoneal injection of Gram-negative bacterial lipopolysaccharide (LPS) solution. Rats in each sepsis group were isolated and mitochondria isolated at the corresponding time points. The mitochondrial membrane potential of rat brain was measured by flow cytometry. The structural damage of mitochondria in various nerve cells was observed by electron microscope. Results: The mitochondrial membrane potential of 3 LPS group was significantly lower than that of the normal control group [(0.5 ± 0.5) vs (1.2 ± 0.6), P <0.05]. The mitochondrial membrane potential in the 3 LPS group was significantly lower than that of the normal control group Significant difference [(1.4 ± 0.7) vs (1.2 ± 0.6), (1.6 ± 0.7) vs (1.2 ± 0.6)]. Compared with the control group, the mitochondrial ultrastructure scores of neurons and astrocytes in 6LPS group were significantly increased (P = 0.000). The mitochondrial ultrastructure scores of microglia and oligodendrocyte in each group were significantly higher than those in control group No significant difference in the control group. CONCLUSION: In the early phase of sepsis, there are reversible function and structural damage of mitochondria in rat brain and functional injury precedes the structural damage. Mitochondrial injury of neurons and astrocytes in sepsis rats is more obvious, Stromal cells and oligodendrocytes have a strong anti-injury ability.
