目的建立铁棒锤高效快速繁殖体系。方法以铁棒锤越冬芽为外植体,用不同的培养基、激素和附加物组合培养。结果培养基MS+2,4-D 2.0 mg/L+NAA0.4 mg/L+活性炭(AC)1.5 g/L+聚乙烯吡咯烷酮(PVP)1 g/L有利愈伤组织的形成;培养基MS+6-BA2.5 mg/L+NAA0.2 mg/L+AC 1.5 g/L+PVP 1 g/L有利于成芽;培养基MS+6-BA3.5 mg/L+NAA0.1 mg/L+2,4-D 0.1 mg/L+AC 1.5 g/L+PVP 1 g/L有利芽增殖;培养基1/2 MS+NAA 0.2 mg/L+AC 1.5 g/L+PVP 1.0 g/L有利于根的生成。结论以种子发芽苗为外植体进行离体培养,可以实现铁棒锤种苗的工业化快速繁殖。
Objective To establish an efficient and rapid propagation system of iron hammer. Methods The overwintering buds were used as explants with different culture media, hormones and additives. Results Media MS + 2,4-D 2.0 mg / L + NAA 0.4 mg / L + Activated carbon 1.5 g / L + polyvinylpyrrolidone (PVP) 1 g / L favorable callus formation; medium MS + 6-BA 2.5 mg / L + NAA 0.2 mg / L + AC 1.5 g / L + PVP 1 g / L was beneficial to bud formation; medium MS + 6- BA 3.5 mg / L + NAA 0.1 mg / L + 2,4-D 0.1 mg / L + AC 1.5 g / L + PVP 1 g / L is conducive to the formation of roots. Conclusion The germination of seedlings as explants in vitro culture can be achieved rapid industrialization of iron seedlings seedlings breeding.
