目的:研究化痰方(二陈汤)对脂代谢紊乱C57BL/6小鼠肝组织中小窝蛋白-1(Cav-1)和磷脂酰肌醇3-激酶(PI3K)/蛋白激酶B(AKT)表达的影响。方法:选取24只SPF级雄性C57BL/6小鼠,用高脂饲料喂养10周建立脂代谢紊乱模型,于第4、10周检测其血脂含量,后随机分为3组:模型组、化痰组及降脂组,每组8只,另选8只小鼠为对照组。药物干预4周后,采用Real-time PCR检测肝组织中Cav-1、PI3K和AKT基因的表达。结果:造模后小鼠血脂[甘油三酯(TG)、总胆固醇(TC)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)]含量均升高,肝组织中Cav-1和AKT表达减少,PI3K表达增加;二陈汤使得模型小鼠血脂含量均有所降低,Cav-1和AKT表达得到回升,而PI3K表达持续增加。结论:化痰方(二陈汤)对脂代谢紊乱小鼠Cav-1/PI3K/AKT信号具有调控作用。
Objective: To study the effect of Huatan Fang (Erchen Decoction) on the expression of caveolin-1 (Cav-1) and phosphatidylinositol 3-kinase (PI3K) / protein kinase B ) Expression. Methods: Twenty-four SPF male C57BL / 6 mice were randomly divided into three groups: model group, phlegm Group and lipid-lowering group, each group of 8, another 8 mice for the control group. Erchenang decoction reduced the blood lipid, the expression of Cav-1 and AKT, and the expression of PI3K in the model mice. Conclusion: Huatan Fang (Erchen Decoction) can regulate the Cav-1 / PI3K / AKT signaling in mice with lipid metabolism disorders.
