骨形态发生蛋白(BMP)为诱导中胚层未分化间充质细胞向成骨细胞方向分化的诱导因子。本实验以猪皮质骨为材料,通过6mol/L脲抽提、分步沉淀、羟基磷灰石层析、凝胶过滤和制备电泳等方法纯化BMP,并通过将BMP植入小鼠骨骼肌诱导成骨测定活性。结果表明,猪BMP的表观分子量小于26000,可能为酸性蛋白质。
Bone morphogenetic protein (BMP) is an inducing factor that induces mesoderm undifferentiated mesenchymal cells to differentiate into osteoblasts. In this study, porcine cortical bone material was used to purify BMP by 6mol / L urea extraction, fractional precipitation, hydroxyapatite chromatography, gel filtration and preparative electrophoresis. BMP was implanted in mouse skeletal muscle Osteogenesis assay activity. The results showed that the apparent molecular weight of pig BMP is less than 26000, which may be acidic protein.
