目的探讨染色体8q24区4个单核苷酸多态性(SNP)rs6983267、rs1447295、rs10090154和rs7837688与北京居民中前列腺癌(PCa)的关联及其基因型和表型的关系。方法收集138名正常对照者和124例PCa患者的血液样本和其遗传、嗜好、临床、膳食习惯等信息。基于高分辨率熔解曲线分析技术(HRM),对124名PCa患者和138名正常对照者的4个SNP(rs6983267、rs1447295、rs10090154和rs7837688)位点进行基因分型。结果 PCa与对照组间rs6983267(55.8%vs 56.8%)、rs1447295(21.5%vs 17.5%)、rs10090154(20.3%vs 16.4%)、rs7837688(19.7%vs 19.3%)各基因型分布差异无统计学意义(P>0.05)。数量性状分析发现,PCa患者中rs7837688的GG基因型携带者饮酒的酒精度数更高(P=0.022)。结论 8q24区rs6983267、rs1447295、rs10090154和rs7837688多态性与北京居民中PCa的易感性可能无相关关系。
Objective To investigate the association of four SNPs rs6983267, rs1447295, rs10090154 and rs7837688 in chromosome 8q24 with the genotypes and phenotypes of Beijing residents with prostate cancer (PCa). Methods The blood samples of 138 normal controls and 124 PCa patients were collected and their genetic, hobby, clinical, and dietary habits were collected. Four SNPs (rs6983267, rs1447295, rs10090154 and rs7837688) sites were genotyped in 124 PCa patients and 138 normal controls based on high-resolution melting curve analysis (HRM). Results There was no significant difference in genotype distribution among rs6983267 (55.8% vs 56.8%), rs1447295 (21.5% vs 17.5%), rs10090154 (20.3% vs 16.4%) and rs7837688 (19.7% vs 19.3% (P> 0.05). Quantitative trait analysis found that alcohol consumption was higher in patients with GG genotype rs7837688 in PCa patients (P = 0.022). Conclusion The polymorphisms of rs6983267, rs1447295, rs10090154 and rs7837688 in 8q24 region may not be related to the susceptibility of PCa in Beijing residents.
