目的观察过氧化物酶体激活物活化受体γ(PPARγ)的配体土槿乙酸(PLAB)联合维甲类X受体α(RXRα)的配体9-顺式维甲酸(9-cisRA)促进白血病细胞凋亡的作用,探讨其作用机制。方法应用透射电子显微镜、琼脂糖凝胶电泳分析HL-60细胞凋亡,RT-PCR检测HL-60细胞Cyclin D1和CDK4 mRNA表达。结果 HL-60细胞经配体联合作用后,电镜下可见明显细胞凋亡的形态学变化,琼脂糖凝胶电泳出现DNA Ladder,Cyc-lin D1及CDK4 mRNA的表达均降低,且联合用药组下降程度明显较单独作用组大(P<0.05)。结论 PLAB联合9-cisRA抑制HL-60细胞生长,其机制与凋亡诱导效应相关。
OBJECTIVE: To observe the effect of ligands of peroxisome proliferator-activated receptor γ (PPARγ), hyaluronic acid (PLAB) and retinoid X receptor α (RXRα) ligand 9-cisRA Promote leukemia cell apoptosis, and explore its mechanism of action. Methods The apoptosis of HL-60 cells was analyzed by transmission electron microscopy and agarose gel electrophoresis. The expressions of Cyclin D1 and CDK4 mRNA in HL-60 cells were detected by RT-PCR. Results The morphological changes of HL-60 cells were observed under electron microscope. The expression of DNA ladder, Cyclin D1 and CDK4 mRNA in the HL-60 cells was decreased by agarose gel electrophoresis, and decreased in combination group The degree of significantly larger than the single role group (P <0.05). Conclusion PLAB combined with 9-cisRA inhibits the growth of HL-60 cells, and its mechanism is related to the induction of apoptosis.
