背景:中枢神经系统中的海马逐渐成为近来研究癫痫发病的重要区域,细胞因子中的白细胞介素2(interleukin-2,IL-2)是一种重要的免疫因子,同时又是中枢神经系统中的一种重要的神经调质。在体及离体的研究证实:脑内存在IL-2及IL-2R,IL-2能影响下丘脑-垂体轴的功能,并能引起行为及脑电图的改变,IL-2与癫痫发病的联系得到广大学者的关注。目的:探讨细胞因子中IL-2与癫痫发病之间的关系。设计:随机对照的实验研究。单位:江汉大学医学与生命科学学院,华中科技大学同济医学院,中国科学院武汉物理与数学研究所波谱与原子分子国家重点实验室。材料:实验于2002-01/12在华中科技大学同济医学院脑研究室完成。健康成年Wistar大鼠52只,雌雄不限,体质量200~250g,实验前均饲养于安静、无强光刺激的环境中。方法:将Wistar大鼠随机分成3组,实验组侧脑室注入马桑内酯(coriarilactone,CL)2μL,生理盐水组注入等量的生理盐水,空白对照组不做任何处理。然后运用免疫组织化学方法,对大鼠海马IL-2的免疫反应性进行分析;同时对实验中大鼠的行为及脑电图进行观察和记录。结果:实验组大鼠侧脑室注射CL过程中即诱发癫痫,且测到痫性脑电图,生理盐水组和空白对照组无癫痫症状且未记录到痫性脑电图;实验组大鼠海马IL-2免疫反应性较生理?
BACKGROUND: The hippocampus in the central nervous system has become an important area of recent studies on the pathogenesis of epilepsy. Interleukin-2 (IL-2) is an important immune factor in cytokines and is also found in the central nervous system An important neuromodulator. In vivo and in vitro studies have confirmed that IL-2 and IL-2R are present in the brain. IL-2 affects the function of the hypothalamic-pituitary axis and causes behavioral and electroencephalographic changes. IL-2 and epilepsy The contact has been the concern of the majority of scholars. Objective: To investigate the relationship between IL-2 and seizure in cytokines. Design: Randomized controlled experimental study. SETTING: School of Medicine and Life Sciences, Jianghan University, Tongji Medical College, Huazhong University of Science and Technology, State Key Laboratory of Magnetic Resonance and Atomic and Molecular Biology, Wuhan Institute of Physics and Mathematics, Chinese Academy of Sciences. MATERIALS: The experiment was performed at Brain Research Laboratory, Tongji Medical College, Huazhong University of Science and Technology from January to December in 2002. 52 healthy adult Wistar rats, male or female, body weight 200 ~ 250g, were kept in the quiet before the experiment, no light stimulus environment. Methods: Wistar rats were randomly divided into 3 groups. The experimental group received intracerebroventricular injection of 2 μL of coriarilactone (CL). The rats in the normal saline group were injected with the same amount of normal saline. The blank control group received no treatment. Immunohistochemistry was used to analyze the immunoreactivity of rat IL-2 in the hippocampus. At the same time, the behavior and EEG of the rats were observed and recorded. Results: Epilepsy was induced in the experimental group by intracerebroventricular injection of CL, and no epileptic symptoms were detected in the epileptic EEG, saline group and blank control group, and no epileptic electroencephalogram was recorded.
