目的研究几种不同种类的铝化合物对神经母细胞瘤细胞(SH-SY5Y)的毒性作用。方法传代培养神经母细胞瘤细胞,分别用乳酸铝(0、2、4、8和16 mmol/L)、氯化铝(0、1、2、4和8 mmol/L)、麦芽酚铝(0、100、200、400和800μmol/L)和柠檬酸铝(0、1、2、4和8 mmol/L)将其染毒24 h。采用CCK-8法检测SH-SY5Y细胞的细胞活力,采用流式细胞术检测SH-SY5Y细胞的凋亡率。结果 (1)与阴性对照组比较,除最低剂量组外,其余剂量的乳酸铝、氯化铝、麦芽酚铝和柠檬酸铝对SH-SY5Y细胞的细胞活性均有明显的抑制作用,且随着染毒浓度的升高,细胞活力明显下降(P<0.05)。(2)流式检测结果显示,与空白对照组相比,各种不同种类的铝化合物均可诱导SH-SY5Y细胞凋亡率增高(P<0.05),并且随着染毒剂量的增加,凋亡率也明显增加(P<0.05)。结论 4种不同种类的铝化合物均可引起神经母细胞瘤细胞的细胞活力降低和凋亡率增加,对SH-SY5Y细胞都具有细胞毒性作用。并且同等浓度时,麦芽酚铝的毒性高于乳酸铝、氯化铝和柠檬酸铝的毒性。
Objective To study the toxic effects of several different kinds of aluminum compounds on neuroblastoma cells (SH-SY5Y). Methods The neuroblastoma cells were subcultured and treated with aluminum lactate (0,2,4,8 and 16 mmol / L), aluminum chloride (0,1,2,4 and 8 mmol / L), maltol aluminum 0, 100, 200, 400 and 800 μmol / L) and aluminum citrate (0, 1, 2, 4 and 8 mmol / L) for 24 h. The cell viability of SH-SY5Y cells was detected by CCK-8 assay. The apoptosis rate of SH-SY5Y cells was detected by flow cytometry. Results (1) Compared with the negative control group, except for the lowest dose group, the other doses of aluminum lactate, aluminum chloride, maltol aluminum and aluminum citrate significantly inhibited the cell viability of SH-SY5Y cells, With the increase of exposure concentration, cell viability decreased significantly (P <0.05). (2) The results of flow cytometry showed that compared with the blank control group, various kinds of aluminum compounds could induce the apoptosis rate of SH-SY5Y cells increased (P <0.05), and with the increase of the dose, Mortality was also significantly increased (P <0.05). CONCLUSION: Four different kinds of aluminum compounds can cause neuroblastoma cells to reduce cell viability and apoptosis rate, and have cytotoxic effect on SH-SY5Y cells. And at the same concentration, maltol aluminum toxicity than aluminum lactate, aluminum chloride and aluminum citrate toxicity.
