建立了反相高效液相色谱法同时测定苹果醋中5种有机酸(酒石酸、苹果酸、乳酸、乙酸、柠檬酸)含量的方法。样品经聚酰胺粉脱色处理后,用C18反相色谱柱进行分离5种有机酸,其中流动相为0.01 mol/L磷酸氢二铵-磷酸溶液,流速为1.0 mL/min,柱温30℃,紫外检测波长210 nm。在选定的色谱条件下,5种有机酸在6 min内能够完全分离,其线性范围为0.001 mg/mL~9.000 mg/mL,标准曲线相关系数均在0.999以上;回收率为96.8%~103.7%,RSD在0.56%~2.11%之间(n=6)。该方法简单快捷、准确、重复性好,可用于苹果醋中有机酸的测定。
A method for the simultaneous determination of five organic acids (tartaric acid, malic acid, lactic acid, acetic acid and citric acid) in apple cider vinegar was established by reversed-phase high performance liquid chromatography. After the sample was decolored with polyamide powder, five organic acids were separated on a C18 reversed-phase column. The mobile phase consisted of 0.01 mol / L diammonium phosphate-phosphoric acid solution, the flow rate was 1.0 mL / min, the column temperature was 30 ℃, UV detection wavelength of 210 nm. Under the selected chromatographic conditions, the five organic acids could be completely separated within 6 min, with a linear range of 0.001 mg / mL to 9.000 mg / mL. The correlation coefficients of the standard curves were all above 0.999. The recoveries ranged from 96.8% to 103.7 %, RSD between 0.56% ~ 2.11% (n = 6). The method is simple and fast, accurate, reproducible, and can be used for the determination of organic acids in apple cider vinegar.
