我们前阶段研究表明维生素B_(12)可减低阿霉素的毒性而不减低其抗瘤活性。本文中,薄板层析和光谱分析表明:两药在体外混合并未产生任何新的化合物。动物实验表明单用阿霉素组小鼠肝匀浆细胞色素P_(450)含量降低,而合用维生素B_(12)则阻止了由阿霉素引起的P_(450)含量的降低,二组差异显著。在使用阿霉素早期,全血谷胱甘肽还原酶活性代偿升高,但后期则趋于耗竭。合用维生素B_(12)减轻由阿霉素引起的这种酶活性耗竭的过程。阿霉素可诱导心肌中脂质过氧化物水平的增高,然而维生素B_(12)可阻止阿霉素所致的心肌脂质过氧化物水平的升高。上述资料也许部份解释了维生素B_(12)对阿霉素的减毒作用机理。
Our previous studies showed that vitamin B_ (12) can reduce the toxicity of doxorubicin without reducing its anti-tumor activity. In this paper, thin-layer chromatography and spectroscopic analysis showed that the two drugs were mixed in vitro and did not produce any new compounds. Animal experiments showed that the single use of doxorubicin in mice liver homogenate cytochrome P 450 content decreased, while the combination of vitamin B 12 prevented the doxorubicin-induced decrease in the content of P 450, the difference between the two groups Significant. In the early use of doxorubicin, whole blood glutathione reductase activity compensatory increase, but tend to be depleted later. Combination of vitamin B_ (12) to reduce the doxorubicin caused by the depletion of this enzyme activity of the process. Doxorubicin induces an increase in lipid peroxides in the myocardium, whereas vitamin B 12 inhibits doxorubicin-induced increases in myocardial lipid peroxides. The above data may explain in part the mechanism of attenuating the effects of vitamin B_ (12) on doxorubicin.
