目的观察莫诺苷对局灶性脑缺血再灌注大鼠皮层CD_(34)表达的影响。方法 45只健康成年雄性Sprague-Dawley大鼠分为假手术组,模型组,莫诺苷小、中、大剂量组,每组9只。线栓法制备大鼠大脑中动脉阻塞30 min后再灌注模型。术后莫诺苷小、中、大剂量组予莫诺苷30 mg/kg、90 mg/kg、270 mg/kg每天灌胃1次。7 d后采用免疫组化(n=6)及Western blotting法(n=3)观察缺血侧皮层CD_(34)表达。结果模型组大鼠患侧皮层CD_(34)表达显著升高,莫诺苷大剂量组较模型组显著升高(F>14.865,P<0.001)。结论莫诺苷可能通过上调局灶性脑缺血再灌注大鼠皮层CD_(34)的表达,促进血管新生。
Objective To observe the effect of morroniside on CD_ (34) expression in the cortex of focal cerebral ischemia-reperfusion in rats. Methods Forty-five healthy male Sprague-Dawley rats were randomly divided into sham-operation group, model group and small, medium and high dose groups of morronisid, with 9 rats in each group. Rat occlusion of middle cerebral artery occluded 30 min after reperfusion by thread plug method. Morroniside was given to small, medium and large doses of morroniside 30 mg / kg, 90 mg / kg and 270 mg / kg once daily. Seven days later, the expression of CD_ (34) in ischemic cortex was observed by immunohistochemistry (n = 6) and Western blotting (n = 3). Results The expression of CD_ (34) in the ipsilateral cortex of rats in model group was significantly higher than that in model group (F> 14.865, P <0.001). Conclusion Morroniside may promote angiogenesis by up-regulating the expression of CD_ (34) in the cortex of focal cerebral ischemia-reperfusion rats.
