目的:比较桑螵蛸生品、制品对小鼠免疫功能和抗氧化能力的影响。方法:对比桑螵蛸生品、制品对巨噬细胞的增殖率和吞噬作用,并测定巨噬细胞所产生的肿瘤坏死因子-α(TNF-α)和一氧化氮(NO)含量;经皮注射环磷酰胺建立小鼠免疫低下模型,测定小鼠血清白细胞介素-2(IL-2)、白细胞介素-4(IL-4)、免疫球蛋白M(Ig M)、免疫球蛋白G(Ig G)含量和胸腺、脾脏指数,体内外实验结合,比较桑螵蛸生品、制品对免疫功能的影响。经皮注射四氧嘧啶造成小鼠氧化损伤模型,测定小鼠血清丙二醛(MDA)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)的含量,并采用蛋白免疫印迹法(western-blotting)检测各组小鼠肝脏组织中SOD的表达,比较桑螵蛸生品、制品对氧化损伤的保护作用。结果:桑螵蛸各给药组均能促进巨噬细胞的增殖,并能提高其吞噬能力,促进巨噬细胞TNF-α和NO的释放,升高免疫低下小鼠血清中IL-2、IL-4、Ig M、Ig G含量和胸腺指数、脾脏指数。提高氧化损伤小鼠血清MDA、SOD、GSH-Px的含量,上调SOD蛋白表达水平。结论:桑螵蛸生品和炮制品均能增强免疫功能,并对氧化损伤具有一定的保护作用,生品的效果优于炮制品。
Objective: To compare the effects of mulberry leaves on the immune function and antioxidant capacity of mice. Methods: The proliferation rate and phagocytosis of macrophages were compared with those of mulberry leaves and the content of tumor necrosis factor-α (TNF-α) and nitric oxide (NO) produced by macrophages were measured. The percutaneous Mice immunosuppressive model was established by injection of cyclophosphamide and the levels of serum interleukin-2 (IL-2), interleukin-4 (IL-4), immunoglobulin M (Ig M), immunoglobulin G (Ig G) content and thymus, spleen index, in vitro and in vivo experiments to compare the mulberry products, products on the immune function. The model of oxidative injury induced by peroxisomes was injected intraperitoneally. The contents of malondialdehyde (MDA), superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) were measured in mice Western blotting was used to detect the expression of SOD in liver tissues of mice in each group. The protective effect of the products on oxidative damage was compared between the two groups. Results: All the mulberry groups could promote the proliferation of macrophages and increase their phagocytic capacity, and promote the release of TNF-α and NO from macrophages, and increase the levels of IL-2, IL in sera of immunocompromised mice -4, Ig M, Ig G content and thymus index, spleen index. Increase the content of MDA, SOD and GSH-Px in serum of oxidative injury mice and up-regulate the expression of SOD protein. Conclusion: Both mullet and processed products can enhance the immune function and have a certain protective effect on oxidative damage. The effect of raw mulberry is better than that of processed products.
