正骨远红外胶贴对慢性软组织损伤动物模型的作用及机理研究

目的:观察正骨远红外胶贴对慢性软组织损伤动物模型的作用并进行初步机理研究。方法:将新西兰大白兔下肢多次击打造成慢性软组织损伤的病理模型,随机分为高剂量组、中剂量组、低剂量组、模型对照组、阳性对照组。以相应的药物贴膏,贴于兔子小腿损伤部位,每天一次,给药6 h,连续给药21 d。在造模后6 h、1d、2 d、3 d、5 d、7 d、14 d、21 d分别进行测量,记录兔子下肢胫骨下周径。于第3 d、7 d、14 d分别取两只,第21 d全部动物按照外表观察评分标准进行评分,然后分别全部处死,取打击部位肌肉组织做病理切片按照以下组织学评分标准进行评分;观察碱性成纤维细胞生长因子的含量变化(免疫组化法)以及组织局部Bax、Bcl-2基因表达;第9 d、16 d、20 d测耳廓微循环的血流速度,记录pu值。第21 d,将兔子全部麻醉,腹腔主动脉取血,检测血清中的IL-1β(放免法)。结果:正骨远红外胶贴能明显抑制软组织损伤兔子的胫骨结节的肿胀;耳廓微循环的血流量没有明显影响;使促凋亡基因BAX在损伤局部的表达增加;bFGF的表达增强。结论正骨远红外胶贴能明显抑制软组织损伤兔子的胫骨结节的肿胀。其机理可能通过促进凋亡基因BAX的表达,促进细胞凋亡,促进bF-GF的表达,抑制血清IL-1β含量升高来发挥其抗炎、抑制损伤、促进修复的作用。 OBJECTIVE: To observe the effect of bone-setting far-infrared adhesive plaster on animal models of chronic soft tissue injury and to study its mechanism. Methods: The pathological models of chronic soft tissue injury caused by multiple beatings of New Zealand white rabbits were randomly divided into high dose group, medium dose group, low dose group, model control group and positive control group. With the corresponding drug paste, affixed to the rabbit leg injury site, once a day, administered 6 h, continuous administration of 21 d. Measurements were made at 6 h, 1 d, 2 d, 3 d, 5 d, 7 d, 14 d, and 21 d after model establishment, respectively. The lower tibia circumference of rabbits was recorded. Two rabbits were sacrificed on day 3, day 7 and day 14, and all animals on the 21 d were scored according to the appearance score. Then all the animals were sacrificed. The muscle tissue of the attack site was made for biopsy and scored according to the following histological grading standards. The changes of basic fibroblast growth factor (BUN) and the expression of Bax and Bcl-2 were observed. The blood flow velocity was measured on the 9th, 16th and 20th days, and the pu value . On the 21st day, all the rabbits were anesthetized and the abdominal aorta was taken for blood test. Serum IL-1β was detected (radioimmunoassay). RESULTS: Posterior foraminotomy significantly inhibited the swelling of tibial tubercle of rabbits damaged by soft tissue. There was no significant effect on the blood flow in the auricle microcirculation. The expression of pro-apoptotic gene BAX increased locally and the expression of bFGF increased. Conclusion The bone glue of far-infrared can significantly inhibit the swelling of the tibial tubercle of rabbits damaged by soft tissues. Its mechanism may exert its anti-inflammatory, inhibit the damage and promote the repair by promoting the expression of apoptosis gene BAX, promoting cell apoptosis, promoting bF-GF expression and inhibiting the increase of serum IL-1β content.
