用PR-HPLC法测定丁公藤注射液有效成分总东莨菪内酯在小鼠血浆中的浓度。色谱条件:SpheriSorb_C18不锈钢柱,流动相为甲醇-水-冰醋酸(38∶62∶0.19),检测波长345nm。血浆样品经酸水解后用氯仿萃取,50℃水浴,氮气吹干,甲醇超声溶解后进样。本法具有快速、简便、灵敏、准确等特点,已经用于总东莨菪内酯血药浓度测定及药代动力学研究,结果满意。
The concentration of total scopoletin in mice plasma was determined by PR-HPLC. Chromatographic conditions: SpheriSorb_C18 stainless steel column, the mobile phase methanol - water - glacial acetic acid (38:62:0.19), detection wavelength 345nm. Plasma samples were acid-hydrolyzed with chloroform extraction, 50 ℃ water bath, nitrogen-dried, dissolved in methanol injection. This method is fast, simple, sensitive and accurate. It has been used in the determination of total scopoletin and its pharmacokinetics. The results are satisfactory.
