目的 寻找检测外周血中肝癌细胞的灵敏方法 ,以便早期发现肝癌患者的血行微转移灶。方法 采用逆转录和巢式多聚酶链式反应及Southern杂交技术 ,将AFPmRNA作为肝癌细胞的特异标志 ,特异扩增检测肝癌患者外周血中微量转移的肝癌细胞。结果 30例正常人和 2 1例肝转移癌均阴性 ,2 4例慢性肝病中有 1例阳性 ;83例肝癌患者有 5 1例阳性 ,阳性率 6 1.5 %。结论 用此方法可以较目前其它检查方法更早发现肝癌患者体内的血行微转移灶。
Objective To find a sensitive method for detecting hepatocellular carcinoma cells in peripheral blood for the early detection of blood micrometastasis in patients with hepatocellular carcinoma. Methods AFP mRNA was used as a specific marker of hepatocellular carcinoma by reverse transcription and nested polymerase chain reaction and Southern blotting. AFP mRNA was specifically amplified to detect the micrometastatic liver cancer cells in peripheral blood of patients with liver cancer. Results 30 cases of normal persons and 21 cases of hepatic metastases were negative, 24 cases of chronic liver disease were positive, 83 cases of liver cancer were positive and the positive rate was 61.5%. Conclusion This method can detect blood micrometastasis in patients with liver cancer earlier than other current examination methods.
