目的研究塞来昔布对人结肠癌细胞系HT-29增殖的抑制作用及其对HT-29细胞微小核糖核酸(mi-RNA)表达谱的影响。方法采用CCK-8比色还原法观察塞来昔布对HT-29细胞增殖的抑制作用;miRNA芯片技术检测塞来昔布作用前后HT-29细胞miRNA的表达变化,实时定量RT-PCR验证其结果。结果塞来昔布对HT-29细胞增殖有显著的抑制作用,且呈剂量时间效应关系,其半数抑制浓度为(237.73±1.65)μmol/L;塞来昔布作用于HT-29细胞48 h后,获得28个与塞来昔布干预相关的miRNAs(P<0.01),其中8个下调,20个上调。结论塞来昔布对miRNA表达谱的影响可能是其抑制HT-29细胞株生长的作用途径之一。
Objective To study the inhibitory effect of celecoxib on the proliferation of human colon carcinoma cell line HT-29 and its effect on the expression of miRNA in HT-29 cells. Methods CCK-8 colorimetric reduction was used to observe the inhibitory effect of celecoxib on the proliferation of HT-29 cells. MiRNA microarray was used to detect the expression of miRNA in HT-29 cells before and after celecoxib treatment. Real-time quantitative RT-PCR result. Results Celecoxib inhibited the proliferation of HT-29 cells in a dose-time-dependent manner. The half-inhibitory concentration was (237.73 ± 1.65) μmol / L and the effect of celecoxib on HT-29 cells was 48 h Twenty-eight miRNAs related to celecoxib intervention were obtained (P <0.01), of which 8 were down-regulated and 20 were up-regulated. Conclusion The effect of celecoxib on the miRNA expression profile may be one of the ways of inhibiting the growth of HT-29 cell line.
