目的研究氧化苦参碱(oxymatrine,OMT)对卵巢癌SKOV3细胞增殖的影响及其可能机制。方法培养SKOV3细胞,应用不同浓度OMT作用不同时间后,应用CCK-8检测细胞增殖的变化;应用Real-Time PCR法检测OMT作用不同时间后,HOTAIR在SKOV3细胞中的表达的变换;应用Lipofect Amine 2000,将HOTAIR过表达慢病毒载体以及其阴性对照转染至SKOV3细胞后,给予OMT,应用CCK-8法检测SKOV3细胞增殖变化,应用Western Blot检测SKOV3细胞Bcl-2以及Bax的蛋白表达含量变化,计算Bcl-2/Bax的比值。结果 OMT明显抑制了SKOV3细胞增殖,并呈时间和剂量依赖;OMT明显降低了HOTAIR在SKOV3细胞中的表达水平,表现为剂量相关;转染HOTAIR过表达慢病毒载体后,OMT抑制增殖的作用被阻断,SKOV3细胞的增殖能力基本恢复,SKOV3细胞中Bcl-2/Bax的比值基本恢复。结论 HOTAIR参与了OMT抑制SKOV3细胞增殖调控过程,其机制可能与调控凋亡相关蛋白Bcl-2和Bax表达相关。
Objective To investigate the effect of oxymatrine (OMT) on the proliferation of ovarian cancer cell line SKOV3 and its possible mechanism. METHODS: SKOV3 cells were cultured and treated with different concentrations of OMT for different time. The changes of cell proliferation were detected by CCK-8. The expression of HOTAIR in SKOV3 cells was detected by Real-Time PCR. Lipofect Amine 2000, the HOTAIR overexpression lentiviral vector and its negative control were transfected into SKOV3 cells, and then given OMT. The proliferation of SKOV3 cells was detected by CCK-8 assay. The protein expression of Bcl-2 and Bax in SKOV3 cells was detected by Western Blot , Calculate the ratio of Bcl-2 / Bax. Results OMT significantly inhibited the proliferation of SKOV3 cells in a dose- and time-dependent manner. OMT significantly decreased the expression of HOTAIR in SKOV3 cells in dose-dependent manner. The effect of OMT on proliferation was inhibited by transfection with HOTAIR over-expressing lentiviral vector The proliferation of SKOV3 cells recovered basically, and the ratio of Bcl-2 / Bax in SKOV3 cells recovered basically. Conclusions HOTAIR is involved in the regulation of proliferation of SKOV3 cells by OMT. The mechanism may be related to the regulation of the expression of Bcl-2 and Bax.
