目的 研究不同转移潜能乳腺癌细胞株中达菲抗原趋化因子受体 (Duffyantigenreceptorforchemokines,DARC)mRNA表达和蛋白质水平的差异 ,探讨DARC与乳腺癌细胞转移潜能的相关性。方法 培养高低转移潜能的人乳腺癌MDA MB 4 35细胞株 ,并将其接种到裸鼠体内 ,以体外培养细胞和裸鼠体内原位异种移植瘤为标本 ,用RT PCR法检测DARCmRNA的表达 ,同时在细胞涂片上 ,用抗Fy3单抗检测DARC的蛋白质水平。结果 高转移潜能MDA MB 4 35细胞的DARC表达显著性地低于低转移潜能MDA MB 4 35细胞 ,并且在接种高转移潜能MDA MB 4 35细胞而形成的裸鼠体内原位移植瘤中的DARC表达也显著性地低于低转移潜能MDA MB 4 35细胞来源的移植瘤。结论 本研究首次揭示趋化因子清除槽DARC的表达差异与人乳腺癌细胞的不同转移潜能有关 ,有望成为抑制乳腺癌转移的新靶点。
Objective To investigate the difference of mRNA and protein expression of Duffyantigen receptor for chemokines (DARC) in different metastatic potential breast cancer cell lines and to explore the correlation between DARC and metastatic potential of breast cancer cells. Methods Human breast cancer cell line MDA MB 4 35 with high and low metastatic potential was cultured and inoculated into nude mice. The in vitro cultured xenografts and nude mice xenografts were used as samples. The expression of DARC mRNA was detected by RT PCR. Meanwhile, on the cell smear, the protein level of DARC was detected with anti-Fy3 monoclonal antibody. Results The DARC expression of high metastatic potential MDA MB 4 35 cells was significantly lower than that of low metastatic MDA MB 4 35 cells, and DARC in orthotopic transplanted tumors inoculated with MDA MB 4 35 cells with high metastatic potential Expression was also significantly lower than that of the low metastatic potential MDA MB 4 35 cell-derived xenografts. Conclusions This study reveals for the first time that the difference in the expression of DARC in chemokine clearance groove is associated with different metastatic potentials of human breast cancer cells and may be a new target for inhibiting breast cancer metastasis.
