目的 :了解人生长激素 (h GH)对于结肠癌细胞有无促增殖和分化作用。 方法 :取对数生长期的人结肠癌细胞株 L OVO,以顺铂和 (或 )不同浓度的 h GH体外培养 ,2 4h后以流式细胞仪测定细胞增殖周期及细胞凋亡等指标。 结果 :同时加顺铂和 h GH培养细胞 ,癌细胞群在 G2 ~ M期发生阻滞 ,细胞凋亡率增加 (P<0 .0 5 ) ,S期细胞数率非常显著地降低 (P<0 .0 1) ;加 h GH 10 0 ng/ m l组 G0 ~ G1 期细胞数率降低 (P<0 .0 5 ) ,S期百分率增高 (P<0 .0 1)。 结论 :L OVO细胞表面可能有 h GH受体 ,h GH在体外作用 L OVO细胞 ,能诱导细胞分化 ,促使静止细胞群进入增殖周期 ,并使处于 DNA合成期的细胞增多 ,为提高肿瘤对细胞周期时相特异性药物的敏感性提供了基础。顺铂能够抑制 h GH促 DNA合成的作用 ,并能提高细胞凋亡率
Objective : To understand the effect of human growth hormone (h GH) on proliferation and differentiation of colon cancer cells. METHODS : Human colon carcinoma cell line L OVO in logarithmic growth phase was cultured in vitro with cisplatin and (or) different concentrations of h GH. Flow cytometry was used to determine cell proliferation cycle and apoptosis. Results: Simultaneously with cisplatin and h GH cultured cells, the cancer cell populations arrested in G2 to M phase, the apoptosis rate increased (P<0.05), and the cell number rate in S phase decreased significantly (P< The percentage of cells in the G0-G1 phase in the h GH 100 ng/ml group was decreased (P < 0.05), and the percentage of S phase was increased (P < 0.01). Conclusions: The L GH receptor may be present on the surface of L OVO cells. h GH acts on L OVO cells in vitro and induces cell differentiation, promotes the stationary cell population to enter the proliferative cycle, and increases the number of cells in the DNA synthesis phase to increase the number of tumor cells. The sensitivity of the phase-specific drug provides the basis. Cisplatin can inhibit the effect of hGH on DNA synthesis and increase the rate of apoptosis
