目的:探讨生脉胶囊对慢性心力衰竭大鼠心肌胶原纤维的影响。方法:将大鼠随机分为假手术组、心衰模型组、生脉胶囊组、卡托普利组、生脉胶囊加卡托普利组,采用腹主动脉缩窄法造成大鼠慢性心力衰竭模型,治疗结束时用Masson染色法测定在心肌室胶原容积分数(CVF),免疫组化法结合图像分析仪进行左心室肌Ⅰ型胶原、Ⅲ型胶原含量的测定。结果:①模型组CVF明显高于假手术组(P<0.01),生脉胶囊组、卡托普利组、生脉胶囊加卡托普利组CVF均低于模型组(P<0.01)。②模型组心肌Ⅰ、Ⅲ型胶原的含量均高于假手术组(P<0.05),生脉胶囊组和卡托普利组Ⅰ、Ⅲ型胶原的含量均低于模型组(P<0.01)。结论:生脉胶囊可降低慢性心力衰竭大鼠心肌胶原容积分数和Ⅰ、Ⅲ型胶原的含量,具有减轻心室重构的作用。
Objective: To investigate the effect of Shengmai Capsule on myocardial collagen fibers in rats with chronic heart failure. METHODS: Rats were randomly divided into sham operation group, heart failure model group, Shengmai capsule group, captopril group, and Shengmai capsule plus captopril group. The rats were subjected to abdominal aorta constriction to induce chronic cardiac arrest in rats. In the failure model, at the end of treatment, the collagen volume fraction (CVF) in the myocardium was measured by Masson staining. The content of type I collagen and type III collagen in the left ventricular myocardium was determined by an immunohistochemical method and an image analyzer. Results: 1The CVF of the model group was significantly higher than that of the sham operation group (P<0.01). The CVF of the Shengmai Capsule group, captopril group and Shengmai Capsule plus captopril group were lower than the model group (P<0.01). 2 The levels of collagen I and III in the model group were higher than those in the sham operation group (P<0.05). The levels of collagen I and III in the Shengmai capsule group and the captopril group were lower than those in the model group (P<0.01). . Conclusion: Shengmai Capsule can reduce myocardial collagen volume fraction and collagen type I and III in rats with chronic heart failure, and it can reduce the effect of ventricular remodeling.
