目的:探讨部分变性高效液相色谱(DHPLC)在检测胃癌组织、血浆中基因突变中的应用。方法:应用DHPLC对39例胃癌组织及血清中p53基因突变进行检测,并测序验证。结果:本研究中p53基因的突变率为21%(8/39);其中肠型胃癌突变率40%(6/15)明显高于弥漫型胃癌8.33%(2/24)(P<0.01);发现既往未见报道突变3例。血清标本中50%(4/8)可扩增出目的片段,但均未检出突变。结论:DHPLC是一项较好的检测胃癌组织中基因突变的筛查方法,在血清中的应用仍需探讨。
Objective: To investigate the application of partial denaturing high performance liquid chromatography (DHPLC) in detecting gastric cancer tissue and plasma gene mutation. Methods: The p53 gene mutation in 39 cases of gastric cancer tissues and serum was detected by DHPLC and sequenced. Results: The mutation rate of p53 gene in this study was 21% (8/39). The mutation rate of intestinal type gastric cancer was 40% (6/15), which was significantly higher than that of diffuse type gastric cancer (8.33%, 2/24) (P <0.01) Found no previous reports of mutations in 3 cases. 50% (4/8) of the serum samples amplified the target fragment, but no mutation was detected. Conclusion: DHPLC is a good screening method for detecting gene mutations in gastric cancer tissues. The application of DHPLC in serum is still to be explored.
