目的探讨人脑胶质瘤及正常脑组织中PTEN及AKT、pAKT、GSK3、pGSK3的表达差异。方法将组织标本分为正常脑组织、低级别胶质瘤、高级别胶质瘤三组,采用Western blot方法检测PTEN、AKT、pAKT、GSK3、pGSK3的表达。结果 PTEN在正常脑组织中表达量最高,在低级别胶质瘤中表达量居中,在高级别胶质瘤中表达量最低(P<0.05)。pAKT及pGSK3则在高级别胶质瘤中表达量最高,低级别胶质瘤中表达量居中,正常脑组织中表达量最低(P<0.05)。AKT及GSK3在三种标本中表达量没有明显差别(P>0.05)。结论 PTEN、pAKT、pGSK3蛋白的表达水平与胶质瘤病理分级具有相关性。随着胶质瘤级别的增高,PTEN蛋白的表达量明显降低,pAKT、pGSK3的表达量明显增高。
Objective To investigate the expressions of PTEN, AKT, pAKT, GSK3 and pGSK3 in human glioma and normal brain tissue. Methods Tissue samples were divided into three groups: normal brain tissue, low grade glioma and high grade glioma. The expressions of PTEN, AKT, pAKT, GSK3 and pGSK3 were detected by Western blot. Results The expression of PTEN was the highest in normal brain tissue, moderate in low grade glioma and lowest in high grade glioma (P <0.05). pAKT and pGSK3 were the highest expression in high-grade glioma, moderate-grade low-grade gliomas, and lowest in normal brain tissues (P <0.05). AKT and GSK3 expression in three specimens no significant difference (P> 0.05). Conclusion The expression of PTEN, pAKT and pGSK3 protein is correlated with the pathological grade of glioma. With the increase of glioma level, the expression of PTEN protein was significantly decreased, and the expression of pAKT and pGSK3 was significantly increased.
