目的:观察轻度低温对脑缺血再灌注后星形胶质细胞活性和组织病理变化的影响。方法:沙土鼠24只,随机分非缺血对照组,缺血对照组和轻度低温组(32°~33C)。夹闭双侧颈总动脉20min 再灌注48h,采用免疫组织化学法检测 GFAP 表达及 HE 染色观察组织病理变化。结果:图象分析显示缺血对照组 GFAP 表达的灰阶值与非缺血对照组比较明显增强(P<0.01),且阳性细胞数增多,胞体肿胀。轻度低温组 GFAP 表达基本恢复到非缺血对照组水平(P<0.05)。缺血对照组海马区部分细胞缺失,有的核固缩;而轻度低温组及非缺血对照组则结构基本正常。结论:轻度低温能明显抑制脑缺血后星形胶质细胞活性,减轻胞体肿胀,并能减轻海马区组织损害程度。
Objective: To observe the effect of mild hypothermia on astrocyte activity and histopathological changes after cerebral ischemia-reperfusion. Methods: 24 gerbils were randomly divided into non-ischemic control group, ischemic control group and mild hypothermia group (32 ° -33C). The bilateral common carotid arteries were occluded for 20 min and then reperfused for 48 h. The expression of GFAP was detected by immunohistochemistry and the histopathological changes were observed by HE staining. Results: The results of image analysis showed that the gray scale value of GFAP expression in ischemic control group was significantly higher than that in non-ischemic control group (P <0.01), and the number of positive cells increased and somatic cell swelling. GFAP expression in mild hypothermia group returned to the level of non-ischemic control group (P <0.05). Ischemia control group, part of the hippocampus cells missing, and some nuclear pyknosis; mild hypothermia and non-ischemic control group, the structure was normal. CONCLUSION: Mild hypothermia can significantly inhibit the activity of astrocytes after cerebral ischemia, reduce the swelling of the body cells and reduce the degree of tissue damage in the hippocampus.
