目的建立高效液相色谱法测定红曲黄酮片中总蒽醌的含量。方法以Waters Sunfire C18(250 mm×4.6 mm,5μm)为色谱柱,甲醇-乙腈-0.1%磷酸(25∶40∶35)为流动相,流速1.0 mL·min-1,柱温35℃,检测波长254 nm。结果色谱峰分离度良好,大黄素在11.50~115.04 ng、大黄酚在12.24~122.40 ng、大黄素甲醚在4.87~48.71 ng内呈良好线性关系。平均加样回收率大黄素为100.6%,大黄酚为101.4%,大黄素甲醚为97.7%。结论该方法简单、快速、重复性好,可用于红曲黄酮片中总蒽醌的质量控制。
OBJECTIVE To establish a HPLC method for the determination of total anthraquinone in monascus flavonoids tablets. Methods Waters Sunfire C18 (250 mm × 4.6 mm, 5 μm) was used as the mobile phase and the mobile phase consisted of methanol-acetonitrile-0.1% phosphoric acid (25:40:35) at a flow rate of 1.0 mL · min- Wavelength 254 nm. Results The resolution of chromatographic peaks was good. Emodin was in the range of 11.50-115.04 ng. The linear relationship was found between 12.24 and 122.40 ng of chrysophanol and 4.87-48.71 ng of physcion. The average recovery was 100.6% for emodin, 101.4% for chrysophanol and 97.7% for physcion. Conclusion The method is simple, rapid and reproducible. It can be used for the quality control of total anthraquinones in.
