目的:探讨天芪益智颗粒对痴呆模型大鼠认知功能及抗氧化能力的影响。方法:建立鹅膏蕈氨酸基底核微量注射AD模型。采用Morris水迷宫法检测大鼠的学习记忆能力变化;尼氏染色法检测海马和大脑皮层神经元病理改变;分光度法检测大鼠脑组织MDA、H_2O_2含量及SOD、GR、CAT活力。结果:与模型组相比,天芪益智颗粒0.63g/kg、0.42g/kg剂量组第5d逃避潜伏期明显降低,0.42g/kg剂量组穿越平台次数明显增加,神经元数量增多;与模型组比较,天芪益智颗粒0.63g/kg、0.42g/kg剂量组显著降低MAD、H_2O_2含量及提高GR活力。结论:天芪益智颗粒具有改善模型大鼠认知功能障碍,保护神经细胞的作用,该作用与其增强抗氧化能力有关。
Objective: To investigate the effect of Tianqi Yizhi Granule on cognitive function and antioxidant capacity in dementia model rats. Methods: The ibotenic acid microinjection into the AD model was established. Morris water maze test was used to detect the change of learning and memory ability. Nissl staining was used to detect the pathological changes of neurons in hippocampus and cerebral cortex. The content of MDA and H_2O_2 and the activities of SOD, GR and CAT in brain of rats were detected by spectrophotometry. Results: Compared with the model group, the escape latency of 0.63g / kg and 0.42g / kg dose group of Tianqi Yizhi Granules was significantly lower than that of the model group, the number of neurons increased significantly in 0.42g / kg dose group, Compared with the control group, Tianqi Yizhi Granules 0.63g / kg, 0.42g / kg dose group significantly reduced MAD, H_2O_2 content and increased GR activity. CONCLUSION: Tianqi Yizhi Granule can improve the cognitive dysfunction and protect nerve cells in rats, which is related to its ability of enhancing anti-oxidation.
