目的:筛选及鉴定出血性中风相关基因。方法:应用差别显示聚合酶链反应(differential display PCR,DD-PCR)技术比较出血性中风病人脑组织出血部位及正常边缘部位的基因表达,并用RT-PCR研究其出血部位及周边正常部位基因的mRNA水平表达情况。结果:经DD-PCR筛选到17个差异片段,其中1个可能为出血性中风相关基因,即人类E2A/HLF融合蛋白mRNA,此基因经RT-PCR证实在出血性中风病人脑组织中高于正常组织。结论:E2A/HLF可能为出血性中风相关基因,在后者发病机制中有一定的作用。
Objective: To screen and identify hemorrhagic stroke related genes. Methods: Differential display PCR (DD-PCR) was used to compare the gene expression of hemorrhage site and normal border area of hemorrhagic stroke patients. RT-PCR was used to study the gene expression of hemorrhage site and surrounding normal mRNA expression level. Results: Seventeen differential fragments were screened by DD-PCR, of which one was likely to be hemorrhagic stroke-associated gene, human E2A / HLF fusion protein mRNA. This gene was confirmed by RT-PCR to be higher than normal in brain tissue of patients with hemorrhagic stroke organization. Conclusion: E2A / HLF may be hemorrhagic stroke-related genes, which may play a role in the pathogenesis of the latter.
