目的:观察角质细胞生长因子(keratinocytegrowthfactor,KGF)对喉癌细胞株Hep-2增殖及细胞外调节信号激酶(extracellularsignal-regulatedkinase,ERK)活性和表达的影响。方法:培养喉癌细胞株Hep-2,MTT比色法及流式细胞仪测定不同浓度KGF对细胞增殖的影响;免疫沉淀法纯化蛋白并用ERK1/2试剂盒测定ERK活性;Western-blot测定p-ERK表达。结果:KGF能显著促进细胞增殖,增强ERK活性及p-ERK表达。结论:KGF可以促使喉癌细胞株Hep-2增殖,其机制与增强ERK活性及p-ERK表达有关。
Objective: To observe the effect of keratinocyte growth factor (KGF) on the proliferation and the activity and expression of extracellular signal-regulated kinase (ERK) in Hep-2 cell line. Methods: The laryngeal carcinoma cell line Hep-2 was cultured, MTT assay and flow cytometry were used to determine the effect of different concentrations of KGF on cell proliferation. The protein was purified by immunoprecipitation and ERK activity was measured by ERK1 / 2 kit. -ERK expression. Results: KGF significantly promoted cell proliferation, enhanced ERK activity and p-ERK expression. Conclusion: KGF can promote the proliferation of laryngeal carcinoma cell line Hep-2, the mechanism of which is related to the enhancement of ERK activity and the expression of p-ERK.
