目的探讨神经生长因子(NGF)对噪声所致听力损伤豚鼠的治疗作用。方法将48只雄性豚鼠随机平均分为4组,即对照组(正常组)、模型组(听力损伤模型组)、穴位注射组、腹腔注射组,除对照组外,另3组豚鼠暴露于声压级100dB(A)的白噪声,然后分别给予NGF穴位注射和腹腔注射,模型组注射生理盐水,对照组不给予任何治疗。于噪声暴露前后以及治疗后用畸变产物耳声发射(DPOAE)技术观察其幅值、信噪比。结果DPOAE幅值在1000,1500,2000,3000,4000,6000,8000Hz频段时,模型组分别为-4.5,-4.3,-6.4,-3.6,-2.3,0.9-11.1;穴位注射组分别为-4.3,0.6,7.9,9.6,11.3,14.6,17.5;DPOAE信噪比在1000,1500,2000,3000,4000,5000,6000,8000Hz频段时,模型组分别为-9.8,-4.7,-1.7,0.2,3.4,6.9,3.7;穴位注射组分别为-1.9,1.7,8.7,11.2,13.1,16.9,20.0;经统计分析,模型组各频段DPOAE幅值和信噪比均明显低于对照组、穴位注射组、腹腔注射组(P<0.01),其中穴位注射组疗效明显好于腹腔注射组;穴位注射组治疗前后DPOAE幅值和信噪比测试结果差异均有统计学意义(P<0.01)。结论外源性NGF穴位注射对噪声所致豚鼠听力损伤有较好效果。
Objective To investigate the therapeutic effect of nerve growth factor (NGF) on guinea pigs with noise-induced hearing impairment. Methods 48 male guinea pigs were randomly divided into 4 groups randomly: control group (normal group), model group (hearing impairment model group), acupoint injection group and intraperitoneal injection group. Except for the control group, the other 3 guinea pigs were exposed to sound The white noise of 100 dB (A) was graded, and then NGF acupoint injection and intraperitoneal injection were given respectively. The model group was injected with saline and the control group was given no treatment. Before and after noise exposure and after treatment, the distortion product otoacoustic emissions (DPOAE) technique was used to observe the amplitude and signal-to-noise ratio. Results When DPOAE amplitude was in the range of 1000, 1500, 2000, 3000, 4000, 6000 and 8000 Hz, the model group was -4.5, -4.3, -6.4, -3.6, -2.3 and 0.9-11.1, 4.3, 0.6, 7.9, 9.6, 11.3, 14.6, and 17.5 respectively; when the signal-to-noise ratio of DPOAE was in the bands of 1000, 1500, 2000, 3000, 4000, 5000, 6000 and 8000 Hz, the model group was -9.8, -4.7, 0.2, 3.4, 6.9, 3.7; acupoint injection group were -1.9,1.7,8.7,11.2,13.1,16.9,20.0; by statistical analysis, DPOAE amplitude and signal to noise ratio in each frequency band were significantly lower than the control group, Acupoint injection group and intraperitoneal injection group (P <0.01). The acupoint injection group was significantly better than that of intraperitoneal injection group. The differences of DPOAE amplitude and signal to noise ratio before and after acupoint injection group were statistically significant (P <0.01) . Conclusion Exogenous NGF injection has good effect on noise-induced hearing impairment in guinea pigs.
